Easter Island drug raises cognition throughout life span in mice

ScienceDaily (June 29, 2012) ? Cognitive skills such as learning and memory diminish with age in everyone, and the drop-off is steepest in Alzheimer's disease. Texas scientists seeking a way to prevent this decline reported exciting results this week with a drug that has Polynesian roots.

The researchers, appointed in the School of Medicine at The University of Texas Health Science Center San Antonio, added rapamycin to the diet of healthy mice throughout the rodents' life span. Rapamycin, a bacterial product first isolated from soil on Easter Island, enhanced learning and memory in young mice and improved these faculties in old mice, the study showed.

"We made the young ones learn, and remember what they learned, better than what is normal," said Veronica Galvan, Ph.D., assistant professor of physiology at the Barshop Institute for Longevity and Aging Studies, part of the UT Health Science Center. "Among the older mice, the ones fed with a diet including rapamycin actually showed an improvement, negating the normal decline that you see in these functions with age."

The drug also lowered anxiety and depressive-like behavior in the mice, Dr. Galvan said. Anxiety and depression are factors that impair human cognitive performance. Lead author Jonathan Halloran conducted scientifically reliable tests to accurately measure these cognitive components in the rodents.

Venturing into the open

Mice are burrowers that prefer tunnels with walls. To observe behavior, Halloran used an elevated maze of tunnels that led to a catwalk. "All of a sudden the mice are in open space," Halloran said. "It's pretty far from the floor for their size, sort of like if a person is hiking and suddenly the trail gets steep. It's pretty far down and not so comfortable."

Mice with less anxiety were more curious to explore the catwalk. "We observed that the mice fed with a diet containing rapamycin spent significantly more time out in the open arms of the catwalk than the animals fed with a regular diet," Halloran said.

The second test measured depressive-like behavior in the rodents. Mice do not like to be held by their tails, which is the way they are moved from cage to cage. Inevitably they struggle to find a way out. "So we can measure how much and how often they struggle as a measure of the motivation they have to get out of an uncomfortable situation," Dr. Galvan said.

Rapamycin acts like an antidepressant

Some mice barely struggle to get free, but if an antidepressant is administered they struggle a lot more. This behavior is very sensitive to the action of antidepressants and is a reliable measure of whether a drug is acting like an antidepressant, Dr. Galvan said.

"We found rapamycin acts like an antidepressant -- it increases the time the mice are trying to get out of the situation," she said. "They don't give up; they struggle more."

The reductions of anxiety and depressive-like behavior in rapamycin-treated mice held true for all ages tested, from 4 months of age (college age in human years) to 12 months old (the equivalent of middle age) to 25 months old (advanced age).

Feel-good chemicals elevated

The researchers measured levels of three "happy, feel-good" neurotransmitters: serotonin, dopamine and norepinephrine. All were significantly augmented in the midbrains of mice treated with rapamycin. "This is super-interesting, something we are going to pursue in the lab," Dr. Galvan said.

Dr. Galvan and her team published research in 2010 showing that rapamycin rescues learning and memory in mice with Alzheimer's-like deficits. The elevation of the three neurotransmitters, which are chemical messengers in the brain, may explain how rapamycin accomplished this, Dr. Galvan said.

Rapamycin is an antifungal agent administered to transplant patients to prevent organ rejection. The drug is named for Rapa Nui, the Polynesian title for Easter Island. This island, 2,000 miles from any population centers, is the famed site of nearly 900 mysterious monolithic statues.

This study became available online June 28 as a manuscript in press in the journal Neuroscience.

Who knew giant multi-touch tables would trigger the next big speed race? Ideum clearly thinks that the PixelSense-based Samsung SUR40 is lagging with that AMD Athlon II X2, because it just rolled out a pair of speed demon 55-inch, 40-point touch surfaces (but not Surfaces) in the Platform and Pro. The Platform has a respectable dual 2.2GHz Core i7 and 8GB of RAM, but it also carries a pair of 256GB solid-state drives in case that museum exhibit app won't load quickly enough. Hopping to the Pro switches to two not quite as speedy 500GB hard drives in standard trim. It more than makes up for this with a quad 3.4GHz Core i7 and NVIDIA's Quadro 600 for the truly stressful projects -- the combination can juggle multiple users and tasks even more smoothly than its MT55 Pro ancestor. Outside of raw speed, picking a table depends mostly on svelteness versus expansion: the Pro has a full-fledged HP tower inside that can drop in SSDs and other upgrades you might fancy, while the Platform is half as thick as a SUR40 (at two inches) and sleeker overall than its big brother. (AP) ? President Barack Obama worried and Republicans celebrated when they first heard the news. But not so fast: In the split-second rush to report the Supreme Court's health care decision Thursday, CNN and Fox News Channel got it wrong.

It was an excruciating test for reporters who were handed a 59-page decision choked with legalese and asked to report its meaning almost instantly.

Bloomberg News and The Associated Press were the first reporting the news ? correctly, at 10:07 a.m. EST ? that the court upheld most of Obama's health care overhaul and a mandate that nearly every American have health insurance. They were followed by Reuters and the SCOTUSblog.

The New York Times made a point of tweeting that reporters and editors were analyzing the decision and would write when they were comfortable that the nuances were correct. The paper didn't tweet the news until 10:20 a.m.

CNN apologized for its error, saying it "regrets that it didn't wait to report out the full and complete opinion" that upheld the mandate requiring virtually all Americans to have health insurance. Fox, however, insisted it was right. "Fox reported the facts, as they came in," said network executive Michael Clemente.

The inaccurate reports were the first ones seen by Obama, who was watching four television monitors outside the Oval Office. White House Counsel Katherine Ruemmler came in moments later with the true story.

It was particularly embarrassing for CNN, which has suffered through one of its worst ratings quarters in several years, primarily due to a paucity of big news. The network eagerly awaited the court's decision, scheduled for 10 a.m., running a "countdown clock" on its screen for hours.

Anchor Wolf Blitzer and reporter Kate Bolduan reported at 10:08 a.m. that the health care law had been struck down, based on a reading of Chief Justice John Roberts' decision that the mandate was not a valid exercise of congressional power under the commerce clause of the Constitution. The screen read: "Supreme Ct. Kills Individual Mandate." The news was tweeted and emailed to the network's followers.

"The court striking down that mandate is a dramatic blow to the president," said CNN reporter John King.

By 10:13 a.m., some doubt had seeped in, and the onscreen headline read: "Supreme Court Rules on Obama Law."

"Let's take a deep breath and see what the justices actually decided," Blitzer said. "It could be more complicated than we originally thought."

Two minutes later, CNN reported the correct decision ? the court had upheld the individual mandate, basing it not on the commerce clause but on Congress' power of taxation. CNN then reported that the entire law had been upheld, with King calling it "a huge, huge victory for President Obama."

On Fox, Bill Hemmer touted the "breaking news" that the individual mandate had been declared unconstitutional. A Twitter account run by Fox anchor Bret Baier's show tweeted the same news. Within two minutes, however, anchor Megyn Kelly was citing SCOTUSblog's report and ordered producers to change an onscreen headline that read: "Supreme Court Finds Health Care Individual Mandate Unconstitutional."

"We're trying to do the best we can," Hemmer said.

The initial report on Baier's Fox show Twitter feed was deleted, followed by the tweet: "Getting word that the individual mandate will survive as a tax ? we are trying to work this out for you ? more to come."

Clemente, Fox executive vice president of news and editorial, was unapologetic. "We gave our viewers the news as it happened," he said. He said Hemmer reported that the mandate was not constitutional under the commerce clause, although the network's reporting gave the impression that the mandate had been fully struck down.

Obama's first news about the decision came from television monitors outside the Oval Office, where the cable channels were reporting that the mandate had been struck down, according to administration officials. Within moments, Ruemmler hurried toward the White House and flashed the president two thumbs up. She explained her reading, and Obama hugged her as Chief of Staff Jack Lew looked on.

Several members of Congress also tweeted incorrect information about the ruling. Republican Rep. Darrell Issa of California called it a "big win for liberty and the Constitution." Florida Republican Rep. Tom Rooney tweeted that it was "great news for the American people, victory for constitution." The politicians later removed the tweets from their Twitter feeds.

The Huffington Post's politics Twitter feed first made the wrong call and corrected itself, saying "we jumped the gun" following the CNN and Fox News reports.

Caution was discussed ahead of time during several planning meetings at The New York Times, said Jim Roberts, assistant managing editor. The paper was willing to take a few extra minutes to make sure it had exactly the right interpretation, he said.

"It's almost stupidly obvious to say, 'We want to be right,' but we want to be right," he said.

The Associated Press ? which delivered the news as a flash, its highest-priority designation ? anticipated a complex decision, and its Washington staff prepared for weeks for the various rulings that might come down, said Kathleen Carroll, the news service's senior vice president and executive editor.

"Our goal, as always: Get the facts and get them out quickly," Carroll said in a memo to the staff. "Once again, the enduring lesson prevails: Expertise and preparation are what make us fast and accurate."

ABC, CBS and NBC all interrupted programming for special reports and all generally got it right. The scene was reminiscent of the Supreme Court's decision that decided the 2000 presidential election between George W. Bush and Al Gore, with reporters standing onscreen, frantically leafing through pages as they tried to read and interpret at the same time.

ABC's Terry Moran had less than a minute to look at the decision before he was talking on the air to anchor George Stephanopoulos and he briefly vamped for time, saying "I'm just taking a quick look at it."

Describing the difficulty of the process in an interview, he said, "you have to be confident enough to say, 'I don't know.'"

In this case, Moran quickly spotted that Roberts had decided the case in agreement with the court's liberal justices, a sign that it was highly unlikely the health care law had been overturned. ABC did bobble one fact initially, incorrectly saying the court's decision was by a 6-3 vote instead of 5-4.

Moran said Thursday's lesson to journalists should be "slow down."

"I actually think the audience is much more interested in understanding than in seeing who finishes first in this case," he said. "In this day and age, there are few true scoops. ... On an event like this, take a breath."

CBS' Jan Crawford made the right call while reading it on the air. NBC's Pete Williams said that "the bottom line here is the Supreme Court has upheld the health care law."

The news media's scrambling quickly became fodder for humor online.

Damon Lindelof, co-creator of the television show "Lost," tweeted that "I am not turning off CNN until they TELL ME GORE WON FLORIDA!"

The website Gawker posted a photoshopped picture of a smiling Obama holding a tablet with CNN's website headlined "Mandate struck down," in a pose reminiscent of the famous photo of President Harry Truman holding up a newspaper with the incorrect "Dewey Defeats Truman" headline.

Analysis: GOP sees tax opening in Obama court win

WASHINGTON (AP) -- For President Barack Obama to turn his Supreme Court victory into a clear-cut political win this fall, he must do something other candidates have failed to do: make voters care that GOP opponent Mitt Romney once embraced the health care policies he now fiercely criticizes.

If Obama can't do that, then Romney may find it easy to fire up conservative activists who despise what they label "Obamacare," while also attracting moderate voters who simply dislike it.

And the Republican will have a new anti-tax argument, thanks to the high court.

On the policy front, Thursday's Supreme Court decision was a huge victory for Obama. It's trickier politically, however, because the court upheld the health care law under Congress' power to tax people, a rationale that surprised many.

In essence, the five-justice majority said, the penalty that Americans will start paying in 2014 if they refuse to obtain health insurance amounts to a tax.

Obama has insisted the fee is not a tax. He also notes that poor people would receive subsidies to buy insurance, which amounts to a tax cut. But his lawyers cited the tax-powers argument in their Supreme Court appearance.

Romney and fellow Republicans immediately launched a line of criticism that often has been potent: Democrats - in this case, Obama and his allies in Congress - are too eager to hike taxes.

"Obamacare raises taxes on the American people by approximately $500 billion," Romney said in his brief remarks.

Sen. Lindsey Graham, R-S.C., told Democratic lawmakers to "stand by your tax increase or stand with us to repeal and replace Obamacare."

Senate Minority Leader Mitch McConnell, R-Ky., said: "The Supreme Court has spoken. This law is a tax."

There's a political problem with that argument, Democrats quickly noted. Romney, while governor of Massachusetts, also required almost every resident to obtain insurance or pay a fee. As former Obama aide Neera Tanden put it, "If you call the mandate a tax increase, then Mitt Romney increased taxes in Massachusetts."

That might inoculate Obama from Romney's tax-hike accusations, if Democrats can make it stick. But Romney's GOP primary opponents repeatedly failed when they tried to wrap "Romneycare" around his neck.

Former Minnesota Gov. Tim Pawlenty reminded Republican voters that Obama "said that he designed Obamacare after Romneycare." Texas Gov. Rick Perry said, "I think Mitt is finally recognizing that the Massachusetts health care plan he passed is a huge problem for him."

Former Sen. Rick Santorum tried hardest of all. He said Romney's health care legacy made him "the worst Republican in the country to put up against Barack Obama."

Romney simply stuck to his criticisms of Obama's health law and avoided detailed discussions of its similarities to his Massachusetts initiative.

Some Democrats think Obama and his well-financed ally groups will do a much better job of painting Romney as a hypocrite or flip-flopper on health care.

Romney's GOP opponents "weren't exactly the most formidable of politicians, and they lacked the resources to really make it an issue on TV," Democratic strategist Doug Thornell said. Romney "is the godfather of the individual mandate and health reform," Thornell added, and thus ill-positioned to lead a fight on the issue.

Jim Kessler, co-founder of the Democratic-leaning group Third Way, said: "The individual mandate that Romney invented in Massachusetts is now a tax. That's a real pickle for Romney."

Republicans have options. They can have surrogates and lawmakers carry the health care battle against Obama, leaving Romney as far removed as possible.

Or it may turn out that voters don't care much about what Romney did as a one-term governor several years ago. That would free him to lead assaults on "Obamacare" with minimum damage.

If that happens, Thursday's court ruling could work against Obama's re-election hopes. Recent AP-GfK polls have found that more Americans oppose the 2010 health care law than support it. Opposition to the "individual mandate" - it would require most people to get insurance or pay a fee - was even deeper in a March poll.

Such findings delight Republican operatives. For Obama, Thursday's ruling was "probably the most damaging of all possible outcomes," GOP strategist Mike McKenna said.

"Identifying the mandate as a tax shears away all of the pretense," McKenna said. "It will energize everyone on the right, even those with deep reservations about Romney."

Another Republican campaign veteran, Terry Holt, said: "Obama might have his law, but the GOP has a cause."

Obama seemed eager to avoid the tax debate Thursday. He did not mention taxes in his 1,200-word speech before cameras.

The president's allies might be more willing to engage. If Republicans insist on calling the health insurance fee a tax, said Tanden, who now heads the Center for American Progress, "then the only person in America who has implemented that policy is Mitt Romney."

People carrying guitars on the street have been a rare sight in Cambodia over the past 10 years.

But now, as the Kingdom?s young population continues to grow creatively, musical-instrument shops are popping up around the capital, with more people interested in learning to play, especially teenagers.

Since its founding a year ago in Phnom Penh?s Boeung Keng Kang I neighbourhood, The Music Arts School, an NGO aimed at promoting arts education, has received as many as 170 students, 70 to 80 per cent of whom have been Cambodians.

Students range from age four to adults, but the majority are between 15 and 16.

?In the past 10 or 20 years, people in the capital didn?t want their children to study music,? says Keth Songva, 28, the director of the Music Arts School. ?I think only a few parents still think like that, but most of them see music as a way to make their children smarter. So they send their children to learn at least one musical instrument.?

The school trains students in both traditional Cambodian and Western instruments, but Songva says students seem to prefer Western instruments to traditional ones.

Guitar is the top preference, followed by piano. The third-favourite musical instrument is the violin, he says.

Although the school?s mission is to make music education accessible, the price of nurturing a nascent talent may seem high to the average Cambodian.

Guitar lessons are US$40 a month for four classes, while piano is $30 and violin $40. Traditional Cambodian instrument classes are slightly cheaper: $24 for a month of kem, Khmer drum or tro.

?Most parents who send their children to music school are upper-class. For lower-class people, this fee will be high for them,? Keth Songva says. ?Music schools have just begun to boom in the past few years, so we really lack music teachers. That?s why the fee is still high.?

For this reason, the school offers scholarships to about 20 per cent of its students who are in financial need.

Men Sonita, 16, spends her time outside of her high school, Phanhasastra International School, learning to play the guitar.

She chose the guitar because it?s more popular than other instruments. She can also carry it easily and play it with or without electricity.

?The guitar fits very well with my favourite Cambodian songs from the 1960s such as Arab Piya, Monkey Dance Monkey and Oh Annie,? Sonita says, referring to the so-called golden age of Cambodian rock and roll.

Sonita?s mother, Nhim Pheakdey, says her daughter had insisted on attending music classes since she was very young.

When Sonita was a bit older, Pheakdey bought her an electronic keyboard and hired a music tutor to teach her at home.

For a short period, Sonita could also play the piano.

?It was her idea to attend music school. It wasn?t our idea. We got her piano lessons, and saw she improved. Once I heard from my foreign friends that children who study music are smarter, we got her the guitar class,? Pheakdey says.

She notes that high schools in Cambodia don?t really have any extra-curricular activities such as sports, so she hopes the music lessons will fill her daughter?s free time and help her do something productive rather than getting into trouble.

To celebrate its first anniversary, the Music Arts School will host a student performance night on July 11.

July 4th in Ocean City | Arts & Entertainment | Atlantic City Weekly

One of South Jersey?s most anticipated spectacles is Independence Day at the Jersey shore. ?

For many, it?s a fixed tradition to be on the beaches and boardwalks for the shore towns? display of fireworks. Natives and vacationers to the Jersey shore can testify that America?s birth is best celebrated in sandals and a bathing suit, accompanied by a tasty boardwalk treat. ?

Ocean City?s Independence Day weekend kicks off with a romantic comedy by Renee Taylor and Joseph Bologna, It Had To Be You, from Friday to Sunday (June 29-July 1) at the Ocean City Repertory Theater. The play is being directed by the founder and producing artistic director Flossi Micciolo. Asbury Avenue is pressing on this summer into its second decade with new In-House Director Eddie Shiffler, appointed by Micciolo. Shiffler was a key component in Ocean City?s 125th Anniversary celebration and will now be helping to reorganize the non-profit organization. ?

FDA panel sees little use for metal-on-metal hips

(AP) ? Government health experts said Thursday there are few reasons to continue using metal-on-metal hip implants, amid growing evidence that the devices can break down early and expose patients to dangerous metallic particles.

The Food and Drug Administration asked its 18-member panel to recommend guidelines for monitoring more than a half-million U.S. patients with metal hip replacements. The devices were originally marketed as a longer-lasting alternative to older ceramic and plastic models. But recent data from the U.K. and other foreign countries suggests they are more likely to deteriorate, exposing patients to higher levels of cobalt, chromium and other metals.

While the FDA has not raised the possibility of removing the devices from the market, most panelists said there were few, if any, cases where they would recommend implanting the devices.

"I do not use metal-on-metal hips, and I can see no reason to do so," said Dr. William Rohr of Mendocino Coast District Hospital, who chaired the meeting.

For decades nearly all orthopedic implants were coated with plastic or ceramic. But in the last 10 years some surgeons began to favor all-metal implants, after laboratory tests suggested the devices would be more resistant to wear and reduce the chances of dislocation.

But recent data gathered from foreign registries shows the devices fail at a higher rate than older implants. That information comes on top of nearly 17,000 reports to the FDA of problems with the implants, which sometimes require invasive surgery to replace them.

The pain and inflammation reported by patients is usually caused by tiny metal particles that seep into the joint, damaging the surrounding tissue and bone. The long-term effects of elevated metal levels in the bloodstream are not clear, though some studies have suggested links to neurological and heart problems.

About 400,000 Americans get a hip replacement each year to relieve pain and restore motion affected by arthritis or injury. Metal hips accounted for about 27 percent of all hip implants in 2010, down from nearly 40 percent in 2008. Doctors have begun turning away from the implants amid several high-profile recalls, including J&J's recall of 93,000 metal hips in 2010.

FDA's experts said Thursday that patients complaining of pain and other symptoms should get regular X-rays and blood testing for metal levels. However, panelists pointed out the problems with the accuracy of blood tests and the difficulties of interpreting the results. There are no standard diagnostic kits for sale that test for chromium and other metals

For patients who are not experiencing pain, panelists said annual X-rays would be sufficient to monitor their implants.

If the FDA ultimately follows the group's advice, U.S. recommendations would be less involved than those already in place overseas.

Earlier this year U.K. regulators recommend that all people who have the implants get yearly blood tests to make sure no dangerous metals are seeping into their bodies.

FDA regulators have suggested they want to take more time to sort out the differences between various implants and patient groups before making recommendations.

"The truth is there are different types of hips and different types of patients," said Dr. William Maisel, FDA's chief scientist for devices, in an interview last week. "Understanding the characteristics of patients who experience adverse events is very important."

Women and overweight people are among the groups that are more likely to have an implant failure.

With little definitive data on U.S. hip implants, the agency has asked manufacturers like Johnson & Johnson, Zimmer Holdings Inc. and Biomet Inc. to conduct long-term, follow-up studies of more than 100 metal-on-metal hips on the U.S. market.

FDA scientists say the studies will help "fill in the blanks" on a number of scientific questions, including the long-term effects of metal particles.

But public health advocates say it could take a decade before that information is available.

"Keeping these metal-on-metal hips on the market for the next five to 10 years while research is conducted is not ethical," said Diana Zuckerman, president of the National Research Center for Women & Families, during a public comment session at the meeting. "If the companies want to sell metal-on-metal hips, they should be required to prove their safety first."
